Avutometinib and Defactinib in Diffuse Gastric Cancer

This trial is testing the combination of two targeted drugs — avutometinib (a MEK inhibitor) and defactinib (a FAK inhibitor) — in people with advanced diffuse-type gastric (stomach) or gastroesophageal junction cancer that has stopped responding to prior treatment. **You may be eligible if...** - You are 18 or older - You have been diagnosed with diffuse-type, poorly cohesive, signet ring cell, or mixed gastric/gastroesophageal junction cancer confirmed by tissue testing - You have already tried at least one prior treatment for advanced disease that included platinum and fluoropyrimidine chemotherapy - Your performance status is good (ECOG 0-1) - Your organ function (blood counts, liver, kidneys) is adequate **You may NOT be eligible if...** - You have not yet received any prior treatment for advanced disease - You have a type of stomach cancer that doesn't match the eligible histological subtypes (unless you carry a CDH1 or RHOA mutation) - You have active uncontrolled infections or severe organ dysfunction - You are pregnant or breastfeeding Talk to your doctor to see if this trial is right for you.
