Loncastuximab Tesirine and Rituximab as Bridging Therapy Before Standard-of-care CAR-T Therapy in Patients With Large B-cell Lymphoma (CORAL)

This study is testing whether using a combination of two drugs — loncastuximab tesirine and rituximab — as a bridge treatment before CAR-T cell therapy can help prepare patients with large B-cell lymphoma (an aggressive blood cancer) for their CAR-T infusion. Bridging therapy helps keep the cancer under control while patients wait for their CAR-T cells to be prepared. **You may be eligible if...** - You are 18 years or older - You have relapsed or refractory large B-cell lymphoma (including DLBCL, transformed follicular lymphoma, or primary mediastinal B-cell lymphoma), or high-grade B-cell lymphoma - You are scheduled to receive commercial CAR-T cell therapy (axicabtagene ciloleucel or lisocabtagene maraleucel) - Your doctor has determined that you need bridging therapy before your CAR-T infusion - You have had at least one prior combination treatment that did not work or stopped working - You have measurable disease on a scan **You may NOT be eligible if...** - You are not planned to receive one of the two specific CAR-T therapies listed - Your doctor does not believe you need bridging therapy - You do not have measurable disease Talk to your doctor to see if this trial is right for you.
